Labile analytes like morphine can be protected by using purified enzymes and short incubation times.

Yes, when using a purified enzyme is possible to perform the hydrolysis directly in the filter or SPE/SLE plates. The use of a purified enzyme avoids protein precipitation and centrifugation steps which are normally necessary when working in presence of high content of protein.

No, Sodium phosphate does not affect columns at levels up to 50 mM and has no significant effect on the overall salt precipitation that can be produced by a methanol mobile phase.

It is very important to maintain the urine:enzyme ratio. These ratios have been established in order to obtain the desired recovery within the indicated time. Decreasing the relative amount of enzyme will require longer incubations and / or lead to lower analyte recoveries.

It is highly recommended that you use the hardest-to-cleave drug in every class as a control. If the hardest-to-cleave analyte is fully hydrolyzed, you may safely assume that all other drugs from the category are also fully hydrolyzed.

BGTurbo’s shelf life is 10 months when kept at 4°C.

BG100’s shelf life is 12 months when kept between 2 to 8°C.

Store the enzyme at the indicated temperature, use only the recommended temperature and  pH during incubation, and mix gently but thoroughly.
