It is very important to maintain the urine:enzyme ratio. These ratios have been established in order to obtain the desired recovery within the indicated time. Decreasing the relative amount of enzyme will require longer incubations and / or lead to lower analyte recoveries.
It is highly recommended that you use the hardest-to-cleave drug in every class as a control. If the hardest-to-cleave analyte is fully hydrolyzed, you may safely assume that all other drugs from the category are also fully hydrolyzed.
